Composite paper-based electrochemical aptasensor for AFB1 detection using Cu-MOF-based CS-PAM hydrogel

Traditional electrochemical methods for AFB1 detection often rely on multiple integrated devices or complex instrumentation systems. To address these limitations, this study presented an innovative composite paper-based aptasensor employing a dual amplification strategy for a signal. By integrating composite paper-based electrodes with Apt/Cu-MOF/CS-PAM nanocomposites on a single platform, the sensor achieves both high sensitivity and portability. The modified paper-based electrodes are better suited to the complex environments encountered in field testing. Cu-MOF can immobilize a large number of aptamers due to its high specific surface area, while the three-dimensional porous network structure of CS-PAM allows for greater loading of Cu-MOF. Upon binding of AFB1 to the aptamers, partial blockage of the channels occurs, restricting effective ion transport pathways and suppressing electrochemical signal output. This enables signal double amplification through a combination of target recognition and mass transfer regulation. Under optimal conditions, the sensor exhibits a detection limit of 0.0003 ng/mL for AFB1, with a linear detection range spanning from 0.001 ng/mL to 100 ng/mL. This work provides a novel strategy for the research and application of paper-based electrochemical sensors.
