We introduce a label-free biosensor based on liquid crystals (LCs) to detect acetamiprid (ACE) in aqueous environments that utilizes an acetamiprid-binding aptamer (ABA) as the recognition element. The sensor ultilized the change in surface topography after the binding of ACE to ABA, which lead to the disruption of LC alginment. The LC orientational transition was observed using a polarized optical microscope (POM). The developed LC-based aptasensor demonstrates a strong linear optical response to ACE across a wide concentration range of 5 pM–70 nM and possesses a low detection limit of 0.69 pM. The biosensor exhibits commendable specificity toward ACE against commonly encountered or structurally similar pesticides. The use of ABA enables the proposed sensor to selectively detect ACE in a mixed pesticide sample. Good recovery rates were obtained when employing our sensor in the detection of ACE spiked in vegetable samples demonstrating the good repeatability and its potential in practical application. This LC-based aptamer sensing approach provides a straightforward, rapid, highly sensitive, selective, and label-free method for analyzing ACE in vegetable samples. © 2024 Elsevier Ltd
