초록

This study introduces a novel pH-responsive polymer derived from polysuccinimide (PSI) that undergoes hydrolysis under high pH conditions, enabling controlled release applications. PSI was applied as a carrier for amoxicillin (AMX) by conjugating AMX to its structure through an aminolysis reaction. The study aims to leverage the stability of PSI at low pH and its hydrolysis activation in the intestinal environment at high pH to protect AMX from degradation in acidic conditions, ensuring its integrity during transit through the digestive system, and triggering release in the intestinal environment, thereby enhancing its efficacy. In vitro tests demonstrated that this carrier prevents AMX release in pH conditions mimicking the acidic environment of the stomach and enables controlled release under conditions reflecting the pH of the intestine. Additionally, it exhibits antibacterial activity against the gram-positive bacterium S. aureus and the gram-negative bacterium E. coli. This innovative approach has the potential to improving antibiotic therapy. © 2024 Elsevier Ltd
